Conditions inducing adventitious embryo and bud from ovule and embryo of Chinese bush cherry (Prununs japonica THUNB.) were studied. For induction of adventitious embryo, MS basal medium containing Gellan gum 0.2 %, sucrose (3, 6, 9 %) and 2, 4-D was used. Ovules collected before flowering did not survive on any medium. In immature embryo, active callus formation was observed on the medium containing 3 % sucrose+15 μM 2, 4-D, while adventitious embryo was induced only 2, 4-D free medium. Although the adventitious embryos were transferred to the hormone-free MS medium after low temperature treatment, they did not grow into plantlet. For induction of adventitious buds, 1/2 MS basal medium containing Gellan gum O.2 %, sucrose 3 % and BA and CPPU (0, 0.2, 2, 10, 20μM), and NAA (0, 0.2,2 μM) was used. From the ovules collected lOdays after flowering, the callus was formed, but adventitious bud was not induced. ln the cotyledons of mature embryo, adventitious buds were formed most actively on the medium with BA 10μM. Addition of NAA suppressed adventitious bud formation. After elongating the adventitious buds on the 1/2 MS medium with 2iP 2.5μM,the base of the shoots were treated with IBA 10 mM and cultured on hormone-free 1/2 MS medium. lOdays after cutting, the shoots started rooting and grew up to the plantlets.
